Target Information
RIPK3 is a member of the receptor-interacting protein (RIP) family of serine/threonine protein kinases, featuring a unique C-terminal domain distinct from other RIP family members.
RIPK3 is mainly localized in the cytoplasm and mitochondria, capable of nucleocytoplasmic shuttling via novel nuclear localization and export signals.
It is part of the tumor necrosis factor (TNF) receptor-I signaling complex and can induce apoptosis and weakly activate NF-κB transcription factor. Caspase-8 prevents RIPK3-dependent necrosis by forming a proteolytically active complex with CFLAR, which is essential for its protective function.
