Target Information
Janus kinase 2 (JAK2)는 특정 사이토카인 수용체 신호전달 경로에 관여하는 단백질 티로신 키나아제입니다. 이 단백질은 JAK2의 촉매 도메인(JH1)을 포함하며, 활성화된 JAKs는 STAT(signal transducers and activators of transcription) 단백질을 인산화시켜 핵으로 이동시키고, 다양한 유전자 전사를 조절합니다.
JAK2는 IFN-γ 수용체 복합체의 신호 개시에 필수적이며, JAK1은 증폭기 역할을 합니다. 일부 연구에서는 Tyk2와 JAK3가 IFN-γ 수용체 복합체 내에서 적절히 위치할 경우 JAK2의 기능을 대체할 수 있다고 제안합니다.
JAK2 변이는 Budd-Chiari 증후군, 진성적혈구증가증(Polycythemia vera), 혈소판증가증(Thrombocythemia 3), 골수섬유증(Myelofibrosis), 급성골수성백혈병(Acute myelogenous leukemia)과 관련이 있습니다.
