Target Information

SIRP alpha (CD172a, signal-regulatory protein alpha) is a receptor-type transmembrane glycoprotein expressed on myeloid-origin cells such as granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells.
It acts as a substrate for activated tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase-coupled signaling pathways.
Binding to CD47 triggers phosphorylation of ITIMs in the cytoplasmic region, recruiting tyrosine phosphatases SHP-1 and SHP-2.
This interaction regulates processes such as inhibition of macrophage phagocytosis and activation of T cells and DCs.
SIRP alpha influences cellular responses to serum, growth factors, insulin, oncogenes, growth hormones, and cell adhesion, playing roles in various physiological and pathological processes.
Cancer cells highly express CD47, which activates SIRP alpha and inhibits macrophage-mediated tumor cell destruction.
