Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and DNA binding to estrogen response elements (ERE). ER regulates gene transcription via coactivator or corepressor recruitment.
Two forms exist—ER alpha and ER beta—encoded by ESR1 and ESR2. Alternative splicing produces multiple ER-alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). ERs are expressed in various tissues, essential for sexual development and reproduction, and implicated in breast cancer, endometrial cancer, and osteoporosis.
