Target Information

Trex1 (ATRIP) is the major human 3’ to 5’ exonuclease required for checkpoint signaling after DNA damage. It is ubiquitously expressed and binds to single-stranded DNA coated with replication protein A at sites of DNA damage, recruiting ATR (ataxia telangiectasia and Rad3-related protein) to these sites. Trex1 is essential for ATR expression.
This gene uses two open reading frames (ORFs):

• The upstream ORF encodes proteins interacting with ATR and localizing to intranuclear foci induced by DNA damage.
• The downstream ORF encodes proteins with 3’ to 5’ exonuclease activity, possibly forming a subunit of human DNA polymerase III.

Multiple transcript variants encoding different isoforms have been identified. Mutations in this gene are associated with Aicardi-Goutières syndrome, chilblain lupus, and Cree encephalitis.
