Product Specific Information

The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a CD8α/β heterodimer or CD8α/α homodimer. CD8a is expressed on most thymocytes, subsets of mature T cells, and NK cells. CD8 binds to MHC class I and associates with p56lck, playing a key role in T-cell development and activation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells at 5 µL (0.06 µg) per test.

Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Use a 780/60 bandpass filter and ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) to minimize polymer interactions.

Compensation Note: Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads (Product # 01-2222-42) compared to stained cells. In such cases, set compensation using cells. Similar effects may be observed with AbC Total Antibody Compensation beads (Product # A10497).

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family involved in signal transduction and T-cell activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α/β) acting as a co-receptor for MHC class I, while CD4 serves as a co-receptor for MHC class II. CD8 is also present as CD8α homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Binding of MHC-I/peptide complexes triggers recruitment of Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs play a key role in pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
