Target Information
SMAD2 (MADH2, MAD2)는 세포 증식, 사멸, 분화 등 다양한 세포 과정을 조절하는 단백질입니다.
이 단백질은 TGF-β 수용체와 상호작용하며, 핵 내로의 이동은 TGF-β 신호전달의 핵심 과정입니다.
ERK1에 의해 MH1 도메인의 칼모듈린 결합 영역의 트레오닌 8이 인산화되면 SMAD2의 전사 활성은 증가하며, 이는 칼모듈린에 의해 음성 조절됩니다.
TGF-β 신호에 반응하여 SMAD2는 수용체에 의해 인산화되고, SARA와의 결합이 해리되어 SMAD4와 결합합니다.
SMAD4와의 복합체는 핵으로 이동하여 표적 프로모터에 결합하고 전사 억제 복합체를 형성합니다.
또한 SMAD2는 activin type 1 receptor kinase에 의해 인산화될 수 있으며, activin 신호를 매개합니다.
대체 스플라이싱된 전사 변이체가 보고되었습니다.
