• For immunohistochemistry (PFA-fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes.
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising about 10% of lymphocyte surface proteins.
It regulates Src-family kinases and plays a crucial role in lymphocyte development and antigen signaling.
Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing transmembrane and cytoplasmic regions.
CD45 isoforms vary in membrane localization and expression depending on cell type and physiological state.
Beyond immunoreceptor signaling, CD45 supports cell survival via integrin-mediated pathways and is involved in apoptosis-related DNA fragmentation.
It is expressed on all human hematopoietic cells except mature red cells and absent in other tissue cell types.
CD45 dysfunction is associated with severe combined immunodeficiency and Hepatitis C.
