Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to constitute the IL-7 receptor (IL-7R), a type I glycoprotein (75–80 kDa). It is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction via tyrosine kinase pathways, including the Jak/STAT pathway, essential for lymphocyte development and T-cell homeostasis. CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs), making it a useful marker for distinguishing Tregs with suppressive activity (Foxp3⁺).

Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Target Information

CD127 (IL-7Rα) is a glycoprotein regulating lymphopoiesis. The functional IL-7 receptor is an alpha/gamma heterodimer, where the gamma(c) chain activates signal transduction and the alpha chain mediates specific signaling. CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells.
Defects in CD127 function are associated with severe combined immunodeficiency (SCID).
