Target Information
DNA methylation, the addition of methyl groups to cytosine bases in CpG dinucleotides, is essential for proper development and gene expression regulation. MBD4, also known as MED1, is a mammalian demethylase enzyme containing a methyl-CpG-binding domain (MBD). It reverts methylated cytosine bases to cytosines by catalyzing the removal of the methyl group as methanol.
MeCP2 and MBD1 repress transcription by binding to methylated DNA, while MBD2 and MBD4 colocalize with heavily methylated satellite DNA and mediate methylation signal functions.
MBD4 shares homology with bacterial base excision repair DNA N-glycosylases/lyases and is sometimes mutated in microsatellite unstable tumors.
