Target Information

CYLD is a 956 amino acid cytoplasmic deubiquitinating enzyme belonging to the ubiquitin carboxy-terminal hydrolases (UCH) family.
It contains three CAP-GLY domains, a proline-rich region, a SH3 binding domain, and a catalytic domain homologous to UCH.
CYLD functions as a tumor suppressor protein influencing the JNK signaling pathway and acts as a negative regulator of TRAF2 and NF-κB signaling.
It also regulates the I-κB kinase pathway and exhibits deubiquitinating activity toward non-Lys-48-linked polyubiquitin chains.
Mutations in CYLD are associated with cylindromatosis, multiple familial trichoepithelioma, and Brooke-Spiegler syndrome.
