Target Information
Focal Adhesion Kinase (FAK)는 125 kDa 비수용체 단백질 티로신 키나아제로, Src의 기질로 작용하며 인테그린 신호전달의 핵심 요소입니다.
FAK는 세포 확산, 분화, 이동, 세포사멸 및 세포주기 G1→S 전이 가속에 중요한 역할을 합니다.
FAK는 중심 촉매 도메인과 C-말단 꼬리를 가지며, 세포외기질과 인테그린 수용체를 통해 세포가 부착하는 접착점에 위치합니다.
인테그린이 피브로넥틴과 결합하면 FAK의 티로신 인산화가 증가합니다.
마우스 NIH3T3 섬유아세포가 피브로넥틴에 부착하면 Grb2 어댑터 단백질과 c-Src PTK가 FAK와 결합하고, ERK2 MAP kinase가 활성화됩니다.
또한, Grb2 SH2 도메인은 티로신 인산화된 FAK에 직접 결합하며, 그 결합 부위는 Tyr925로 확인되었습니다.
FAK 유전자는 여러 전사 변이체를 가지며, 그중 일부만 전체 길이 형태로 확인되었습니다.
