Target Information
Mitochondrial DNA polymerase is a heterotrimeric enzyme complex composed of a homodimer of accessory subunits and a catalytic subunit. The POLG gene encodes the catalytic subunit of this enzyme, which contains a polyglutamine tract near its N-terminus that may be polymorphic.
Mutations in this gene are associated with several mitochondrial disorders, including:
- Progressive external ophthalmoplegia with mitochondrial DNA deletions 1 (PEOA1)
- Sensory ataxic neuropathy dysarthria and ophthalmoparesis (SANDO)
- Alpers-Huttenlocher syndrome (AHS)
- Mitochondrial neurogastrointestinal encephalopathy syndrome (MNGIE)
