Product Specific Information

1 µg/mL of MA5-45323 was sufficient for detection of GRP78 in 20 µg of HEK-293 lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody. Detects approximately 78 kDa.

Target Information

GRP78 is a 78 kDa glucose-regulated protein belonging to the HSP70 family. It resides in the endoplasmic reticulum (ER) and associates with newly synthesized secretory and membrane proteins. GRP78 also interacts with mutant or misfolded proteins, targeting them for degradation.
The conserved C-terminal sequence Lys-Asp-Glu-Leu (KDEL) enables ER retention via the KDEL receptor. GRP78 assists in protein folding and degradation of misfolded proteins through interaction with DNAJC10, maintaining cellular homeostasis and preventing apoptosis.
GRP78 levels serve as biomarkers for hypoglycemia and have neuroprotective roles under oxidative stress. Reduced GRP78 expression is observed in Alzheimer’s disease and is linked to PS1 gene mutations. Its induction is also associated with antitumor drug resistance.
