Target Information
Specialized cells at the midline, which separates the left and right halves of the CNS, play key roles in directing growth cone behavior. In vertebrates, insects, and C. elegans, midline cells produce guidance cues such as nectins and slit, acting as attractants and repellents. Robo proteins serve as axon guidance receptors and belong to a conserved subfamily of Ig superfamily proteins. Robo acts as a receptor for Slit, functioning cell-autonomously. Non-crossing axons express high levels of Robo, whereas crossing axons increase Robo expression after crossing the midline. Robo1 and Robo2 are human homologs of the Drosophila Roundabout protein; Robo1 is homologous to C. elegans sax3 and Robo2 to zebrafish astray.
