Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1. PD-L2 is primarily expressed in dendritic cells, but its expression can be induced in various immune and non-immune cells depending on the microenvironment.

Expression is upregulated by cytokines such as IL-4, TNF-α, and IFN-γ. Compared to PD-L1, PD-L2 shows 2–6 times higher affinity for PD-1. PD-1 and its ligands function as inhibitory immune checkpoint molecules, modulating immune responses by inhibiting T cell activation and cytokine secretion (e.g., IFN-γ, IL-10).

Recent studies indicate that PD-L2 also interacts with a second receptor, repulsive guidance molecule b (RGMb), which participates in respiratory tolerance regulation through BMP and/or neogenin pathways. The native human PD-L2 protein includes a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
