Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCam-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) in peripheral lymphoid tissues.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤0.2 µg/test)

A test is defined as the antibody amount staining a cell sample in 100 µL volume. Cell number may vary (10⁵–10⁸ cells/test). Titrate antibody for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample (100 µL of cells, 10³–10⁸ cells) before combining with cells.
  • Required for antibody cocktails and single-color controls.

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures and minimized spillover/spread.

Laser: 561 nm (Yellow)

Internal testing shows NovaFluor Yellow 660 non-specifically stains B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 mice.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It consists of:

• Aminoterminal C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on all leukocyte classes, including lymphocytes (except some memory T cells), monocytes, and polymorphonuclear cells.
