Product Specific Information
The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express CD44, and peripheral B and T cells can upregulate its expression. CD44 functions as an adhesion molecule through binding to hyaluronate, an extracellular matrix component.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse bone marrow cells and splenocytes at ≤0.6 µg per test (100 µL final volume). Recommended to titrate for optimal assay performance.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
- Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.

Target Information
CD44 is a 100 kDa type I transmembrane glycoprotein widely expressed on human leukocytes, brain white matter, and epithelial cells of the intestine and breast. It serves as a receptor for hyaluronic acid (HA) and is involved in cell-cell interactions, adhesion, and migration. CD44 participates in lymphocyte activation, recirculation, and homing, and its expression may be upregulated in certain carcinomas, potentially linked to metastasis.
CD44 expression is found in hematopoietic and non-hematopoietic cells, epithelial tissues, and filopodia in cultured keratinocytes. Bone marrow myeloid cells and memory T cells express CD44 at high levels. Transcripts undergo complex alternative splicing, producing functionally distinct isoforms.
Associated Diseases: Superficial keratitis, lichen sclerosus, and possibly tumor metastasis formation.
