Product Specific Information

The CD28.2 monoclonal antibody reacts with human CD28, a 44 kDa homodimer expressed on thymocytes, mature T cells, and plasma cells. CD28 acts as a ligand for CD80 (B7-1) and CD86 (B7-2), providing potent co-stimulation for T cell activation. Signaling through CD28 enhances IL-2 expression and cytotoxicity in CD3-activated T cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per 100 µL cell sample (10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended detection with a 610/20 bandpass filter. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57). Performance after fixation may vary by clone.

Target Information

CD28 is a 44 kDa disulfide-linked homodimeric T cell surface glycoprotein of the immunoglobulin superfamily. It is a critical co-stimulatory receptor providing the second activation signal for T cells via binding to CD80 and CD86 on antigen-presenting cells. CD28 also prevents T cell anergy and premature apoptosis. In mice, CD28 is expressed on all CD4⁺ and CD8⁺ T cells, with expression varying by thymocyte maturation stage.
Diseases associated with CD28 dysfunction include mycosis fungoides and Sezary’s disease.
