Target Information
ZAP-70 is a cytosolic protein tyrosine kinase belonging to the Syk family. It is expressed exclusively in T-cells and natural killer cells and is required for T-cell receptor activation. Upon T cell antigen receptor (TCR) engagement, ZAP-70 is phosphorylated on tyrosines 292, 315, and 319 in the interdomain B region between the SH2 and kinase domains. Phosphorylation of Tyr315 (Vav binding site) and Tyr319 (Lck binding site) enhances ZAP-70 function in lymphocyte signaling, whereas phosphorylation of Tyr292 terminates activation and attenuates signaling.
