Description:
The monoclonal antibody 29A1.4 recognizes mouse NKp46 (CD335), a member of the natural cytotoxicity receptor (NCR) family. CD335 is a glycoprotein with two Ig-like domains and a short cytoplasmic tail, expressed exclusively on NK cells (including immature NK cells, DX5- CD3-). It allows discrimination between NK and NKT cells (NKp46+, CD3-).
Staining with 29A1.4 is not strain-specific and has been shown in C57Bl/6, SJL, CBA/CA, and BALB/C strains. NKp46 plays a role in NK cell-mediated lysis of tumor and pathogen-infected cells.

The 29A1.4 antibody activates NK cells in vitro but does not deplete them in vivo.
Applications reported include flow cytometric analysis of mouse splenocytes, recommended at ≤1 µg/test. Optimal titration is advised per assay conditions.

Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm lasers and emits at 607 nm. A 610/20 band-pass filter (equivalent to PE-Texas Red®) is recommended. Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal signal loss. Clone-specific performance should be verified empirically.

Target Information

NKp46 (CD335, NCR1) is a cytotoxicity-activating receptor contributing to NK cell-mediated tumor cell lysis. It consists of two Ig-like domains similar to those in LIR and KIR receptor families.
