Cellular Localization Cell membrane; Single-pass type I membrane protein. Upon NRG1 treatment, the activated receptor is internalized. The ERBB4 intracellular domain (E4ICD) translocates to the nucleus and mitochondrion. E4ICD2 is predominantly nuclear and colocalizes with YAP1.

Background

This gene belongs to the Tyr protein kinase family and the epidermal growth factor receptor subfamily. It encodes a single-pass type I membrane protein with multiple cysteine-rich domains, a transmembrane domain, a tyrosine kinase domain, a phosphatidylinositol-3 kinase binding site, and a PDZ domain binding motif. The protein binds to and is activated by neuregulins and other factors, inducing various cellular responses including mitogenesis and differentiation. Proteolytic processing releases cytoplasmic and extracellular fragments. Mutations in this gene are associated with cancer. Multiple alternatively spliced variants exist, encoding different isoforms, though not all are fully characterized. (RefSeq, Jul 2008)
