Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- For formalin-fixed tissues, heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at room temperature for 20 min.
Target Information
Calponin is a thin filament-associated protein involved in smooth muscle contraction regulation. It binds to actin, calmodulin, troponin C, and tropomyosin.
- Expressed in smooth muscle and tissues with smooth muscle components.
- Two isoforms: 34 kDa and 29 kDa; the 29 kDa form mainly in urogenital tract muscle.
- Also expressed in myoepithelial cells from benign and malignant breast lesions.
- Stains smooth muscle, myoepithelial cells, myofibroblasts, keratinocytes, and nerve fibers.
- Useful for differentiating breast collagenous spherulosis (positive) from adenoid cystic carcinoma.
- Adenoid cystic carcinoma in salivary gland tumors is calponin positive.
