This antibody recognizes cyclin D1, an ubiquitously expressed 33 kDa intracellular protein that migrates as a 36 kDa band under reducing SDS-PAGE conditions.
Flow cytometry: Intracellular staining requires membrane permeabilization.

Target Information

Cyclin D1 (PRAD-1, bcl-1) is a key regulator of the cell cycle, functioning with CDK4 and/or CDK6 to phosphorylate the Rb protein. It acts as a proto-oncogene and is overexpressed in various human neoplasms, including mantle cell lymphomas (MCL). Cyclin D1 regulates protein phosphorylation, mammary epithelial proliferation, and adipocyte differentiation.

The CCND1 gene encoding cyclin D1 is located on chromosome 11. Cyclin D1 is both cytoplasmic and nuclear, synthesized during G1 phase, and forms complexes with CDK4 or CDK6 in response to growth factors. These complexes inactivate Rb via phosphorylation and modulate CDK inhibitors p21Cip1 and p27Kip1. Without stimulation, Cyclin D1 undergoes ubiquitin-mediated degradation, contributing to G1/S phase arrest.

Cyclin D1 belongs to the conserved cyclin family, characterized by periodic protein abundance during the cell cycle. It serves as a regulatory subunit for CDK4/6, essential for G1/S transition, and interacts with Rb. Alterations in Cyclin D1 expression are linked to tumorigenesis and are significant in cancer research and cell cycle studies.
