Target Information
CREB (Cyclic AMP response element binding protein) is a 43 kDa basic/leucine zipper transcription factor that binds to the cyclic AMP response element (CRE) and activates transcription in response to various extracellular signals such as neurotransmitters, hormones, membrane depolarization, and growth and neurotrophic factors.
Activation of CREB depends on phosphorylation at serine 133, which occurs via p44/42 MAP kinase, p90RSK, p38 MAP kinase, and MSK1.
Although CREB binds DNA regardless of phosphorylation, only the phosphorylated form acts as a transcription factor. CREB binding protein (CBP), a transcriptional coactivator, interacts with CREB at the serine 133 region.
CREB is involved in diverse cellular processes including circadian rhythm synchronization, adipocyte differentiation, and learning and memory. In humans, the gene is located on chromosome 2q.
Dysfunction of CREB is associated with diseases such as Alzheimer’s disease, histiocytoma, and soft tissue melanoma.
