Target Information
Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), referred to as A-to-I RNA editing. This process can affect gene expression and function by altering codons, splice sites, RNA stability, and RNA structure-dependent activities such as microRNA production and protein-RNA interactions.
ADAR can edit both viral and cellular RNAs, either at multiple sites (hyper-editing) or specific sites (site-specific editing). Its cellular RNA substrates include:
- Bladder cancer-associated protein (BLCAP)
- Neurotransmitter receptors for glutamate (GRIA2), serotonin (HTR2C), and GABA receptor (GABRA3)
Site-specific RNA editing of these transcripts leads to amino acid substitutions that alter protein function.
Viral RNA substrates include:
- Hepatitis C virus (HCV)
- Vesicular stomatitis virus (VSV)
- Measles virus (MV)
- Hepatitis delta virus (HDV)
- Human immunodeficiency virus type 1 (HIV-1)
ADAR1 exhibits proviral or antiviral effects depending on the virus and mechanism:
- Proviral: HDV, MV, VSV, HIV-1
- Antiviral: HCV
It can modulate viral replication via editing-dependent or editing-independent mechanisms. For example:
- Inhibits HCV replication through RNA editing
- Enhances MV, VSV, and HIV-1 replication by suppressing EIF2AK2/PKR activation
- Stimulates HIV-1 particle release and infectivity through editing in the 5′UTR and coding sequences of Rev and Tat
- Enhances HDV replication via A-to-I editing that converts a stop codon (amber) to a tryptophan codon, allowing synthesis of the large delta antigen (L-HDAg) required for viral assembly
- High ADAR1 expression can inhibit HDV replication
