Target Information

Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), referred to as A-to-I RNA editing.
This process can affect gene expression and function by:

  • Modifying codons and amino acid sequences in proteins
  • Altering splice site recognition sequences in pre-mRNA
  • Changing RNA stability and nuclease recognition
  • Influencing RNA virus genome replication
  • Regulating microRNA production and RNA-protein interactions

ADAR can edit both viral and cellular RNAs, either at multiple sites (hyper-editing) or specific sites (site-specific editing).
Its cellular RNA substrates include:

  • BLCAP (bladder cancer-associated protein)
  • GRIA2 (glutamate receptor)
  • HTR2C (serotonin receptor)
  • GABRA3 (GABA receptor)

Viral RNA substrates include:

  • Hepatitis C virus (HCV)
  • Vesicular stomatitis virus (VSV)
  • Measles virus (MV)
  • Hepatitis delta virus (HDV)
  • HIV-1

ADAR1 exhibits both proviral and antiviral effects, depending on virus type and mechanism (editing-dependent or independent).
It can impair HCV replication via RNA editing and enhance replication of MV, VSV, and HIV-1 through suppression of EIF2AK2/PKR activation.
In HDV, ADAR1 promotes viral replication via A-to-I editing at the amber/W site, converting a stop codon to a tryptophan codon, allowing synthesis of the large delta antigen (L-HDAg).
However, high levels of ADAR1 inhibit HDV replication.
