Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. p21 plays a key role in the cellular response to genotoxic stress as a major transcriptional target of p53. In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking cell cycle progression from G1 to S phase or G2 to M phase after DNA damage to allow DNA repair. In the cytoplasm, p21 exhibits anti-apoptotic activity by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK. p21 has a dual role in carcinogenesis, functioning both as a tumor suppressor and as an oncogene depending on context.
