Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell-specific surface glycoprotein. It belongs to the immunoglobulin superfamily and is constitutively expressed on most peripheral blood T lymphocytes.
CD28 acts as a critical T cell co-stimulatory receptor by binding to CD80 and CD86 on antigen-presenting cells, providing the necessary second activation signal. It also prevents T cells from entering an anergic or apoptotic state.
In mice, CD28 is expressed on all CD4⁺ and CD8⁺ T cells in peripheral lymphoid organs and blood. Expression is highest on immature thymocytes (CD3⁻, CD8⁺, CD4⁺8⁺, and CD4⁻8⁻ cells expressing α/β or γ/δ TCR). Mature thymocytes express CD28 at two- to fourfold lower levels.
CD28 dysfunction has been associated with diseases such as mycosis fungoides and Sézary’s disease.
