Target Information
TP63 (tumor protein 63) acts as a sequence-specific DNA binding transcriptional activator or repressor. Isoforms contain varying transactivation and inhibitory domains, showing isoform-specific activity. Isoform 2 activates RIPK4 transcription. TP63 may cooperate with TP73/p73 for p53/TP53-dependent apoptosis in response to genotoxic stress and oncogene activation. It is involved in Notch signaling via JAG1 and JAG2 induction and plays a role in epithelial morphogenesis regulation. The ratio of DeltaN-type and TA-type isoforms influences epithelial stem cell maintenance and stratification initiation. TP63 is required for limb formation from the apical ectodermal ridge and can activate p21 promoter transcription. Mutations in TP63 are associated with several syndromes, including acro-dermato-ungual-lacrimal-tooth syndrome, ectodermal dysplasia variants, and orofacial cleft 8.
