Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is breast ductal carcinoma.
Target Information
Calpastatin is an endogenous inhibitor of calpain, a calcium-dependent cysteine protease.
It consists of an N-terminal domain L and four repetitive calpain-inhibition domains (domains 1–4).
Calpastatin is involved in proteolysis of amyloid precursor protein and in various membrane fusion events such as neural vesicle exocytosis and platelet and red-cell aggregation.
It may also influence expression levels of structural or regulatory proteins.
