The MEM-15 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes via surface CD14.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.05 µg)/test.
A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Invitrogen LIVE/DEAD Fixable Dead Cell stains are recommended for NovaFluor dyes.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation over CellBlox.
Use 5 µL per sample for optimal staining.
For single-color controls, use 5 µL of CellBlox Blocking Buffer per 100 µL of sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton technology, utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures with reduced spillover and spread.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils.
It functions as a multifunctional lipopolysaccharide receptor and can be released into serum as soluble sCD14.
CD14 binds LPS via LBP and contributes to immune responses, apoptosis recognition, and inflammatory regulation.
Alternative splicing produces multiple transcript variants encoding the same isoform.

Diseases associated with CD14 dysfunction include Mycobacterium chelonae infection and Croup.
