Product Specific Information

MA1-13042 detects STAT3 from human, mouse, monkey, and rat samples.
It has been validated for Western blot, ChIP, immunofluorescence, immunohistochemistry (paraffin), and immunoprecipitation.
By Western blot, this antibody detects an 88 kDa protein corresponding to STAT3.
Positive control: HepG2 cells.

The antigen is a recombinant protein corresponding to residues 655–770 of human STAT3.
This antibody replaces a previous version sold as part of the Thermo Scientific Cellomics High Content Screening Kit.
It is recommended for high-content cell-based assays and has been thoroughly tested for cellular immunofluorescence (IF) applications.
Optimization, including selection of the most appropriate fluorescent DyLight-conjugated secondary antibody, may be required for specific assays.

Target Information

STAT3 is a member of the STAT (signal transducers and activators of transcription) family.
It is phosphorylated by receptor-associated kinases, translocates to the nucleus, and acts as a transcription factor in response to cytokines and growth factors.
Coactivators such as CREB-binding protein are necessary for transcriptional activation.
STAT3 can be activated by interferons (alpha, gamma), EGF, PDGF, and IL6.
Phosphorylation at tyrosine 705 by JAK1 and JAK2 is essential for dimer formation, nuclear translocation, and DNA binding.
Located on chromosome 17q in humans, STAT3 mediates gene expression in response to cell stimuli, influencing cell growth and apoptosis.
Interactions include Rac1 (activator) and PIAS3 (specific inhibitor).
Three alternatively spliced transcript variants encode distinct isoforms.
Knockout of STAT3 in mice results in early embryonic lethality.
