Target Information

Janus kinase 2 (JAK2) is a protein tyrosine kinase involved in cytokine receptor signaling and is mutated in polycythemia vera.
This kinase contains the catalytic domain (JH1) of JAK2. Activation of JAKs leads to phosphorylation of STAT proteins, which dimerize and translocate to the nucleus to regulate gene transcription, including interferon-stimulated genes.
JAK2 is required for IFN gamma-receptor complex initiation, while JAK1 acts as an amplifier.
Mutations in JAK2 are associated with Budd-Chiari syndrome, Polycythemia vera, Thrombocythemia 3, Myelofibrosis, and Acute myelogenous leukemia.
