Target Information
Synaptotagmin 1 is a synaptic vesicle membrane glycoprotein widely expressed throughout the central nervous system. It functions as a Ca²⁺ sensor regulating exocytosis and neurotransmitter release. Synaptotagmin’s Ca²⁺-mediated binding to SNAP25 is essential for Ca²⁺-dependent membrane fusion. Specific residues in the C2B binding domain influence endocytic rate and vesicle size. It also binds acidic phospholipids requiring both an acidic head group and diacyl backbone. Ca²⁺-dependent interactions occur with receptors for activated protein kinase C, while Ca²⁺-independent binding occurs with neurexins, syntaxin, and AP2. Dysfunctions in Synaptotagmin 1 are linked to food-borne and infant botulism.
