Target Information
FGF2 (fibroblast growth factor basic, FGFb) is a member of the fibroblast growth factor family that interacts with high-affinity transmembrane receptors to regulate cell proliferation and neovascularization.
It exists as five isoforms with distinct intracellular localizations and functions:
- The 18 kDa isoform is cytosolic and acts through cell surface receptors.
- The 22, 22.5, 24, and 34 kDa isoforms are nuclear and may signal independently of transmembrane receptor pathways.
In humans, the FGF2 gene is located on chromosome 4q and plays roles in limb and nervous system development, wound healing, and tumor growth.
FGF2 mRNA contains multiple polyadenylation sites and is alternatively translated from non-AUG and AUG initiation codons, producing five isoforms with distinct properties.
CUG-initiated isoforms are nuclear (intracrine signaling), while AUG-initiated forms are cytosolic (paracrine and autocrine signaling).
FGF2 dysfunction has been associated with Kaposi Sarcoma and corneal neovascularization.
