Target Information
ATG5 (Autophagy Related 5)는 자가포식(autophagy)에 중요한 요소로, 세포사(apoptosis) 과정에도 관여합니다. 또한 산화 손상 후 미토콘드리아 품질 관리, 선천적 항바이러스 면역 반응의 음성 조절, 림프구 발달 및 증식, MHC II 항원 제시, 지방세포 분화 등의 세포 과정에 관여합니다.
ATG12와의 결합 후 자가포좀 형성에 참여하며, FADD 단백질과 상호작용하여 자가포식성 세포사에 기여합니다. ATG5-ATG12 복합체는 자가포좀 형성 직전 또는 직후에 막으로부터 분리되는 컵 모양의 격리막을 형성합니다.
또한 RIG-I 및 VISA(IPS-1)와 같은 선천면역 단백질과 결합하여 제1형 인터페론 생성을 억제하고 바이러스 복제를 허용합니다. ATG5 기능 이상은 소뇌실조증(spinocerebellar ataxia)과 관련이 있습니다.
