Product Description

E. coli O157:H7 is a rod-shaped, gram-negative bacterium known for producing Shiga-like toxins that can cause severe symptoms including hemorrhagic colitis and hemolytic uremic syndrome. Infection typically results from consumption of undercooked meat, untreated water, or raw milk.

Norgen’s Escherichia coli O157:H7 Quantified Bacterial DNA Standards (STX1 and STX2 genes) are cloned fragments of the Shiga-like toxin regions purified using Norgen’s sample preparation technology. They include partial sequences of STX-1 subunit A and STX-2 subunit A genes. The purified DNA is quantified using spectrophotometry, gel densitometry, and real-time PCR. Intended for use as positive controls or PCR quantification standards for E. coli O157:H7. Compatible with Norgen Biotek’s E. coli O157:H7 TaqMan PCR Detection Kit (Cat. TM41350) and Probe/Primer & Control Set (Cat. TM41310).

2 μL of Norgen’s E. coli O157:H7 Quantified Bacterial DNA Standards (STX1 and STX2 genes) and serial dilutions (colored curves) were used in a 20 μL E. coli O157:H7-specific PCR reaction using Norgen’s 2X PCR Master Mix (#28007). Using STX-1 and STX-2 specific TaqMan Primers/Probes, separate reactions were set up for STX1 and STX2 gene detection. Real-time PCR reactions were performed on a Bio-Rad CFX Real-Time PCR System. No-template controls with water were also tested (black curves). Consistent amplification was observed down to 20 copies/μL.
