Target Information
Rho GTPases, activated by specific guanine-nucleotide exchange factors (GEFs), are essential for various cellular functions. TEM4, encoding a 1510-amino-acid protein, contains a RhoGEF-specific Dbl homology (DH) domain but lacks the typical pleckstrin homology domain. TEM4 acts as a Rho-specific GEF with unique structural and regulatory features and is predominantly expressed in the heart. It links tyrosine kinase signaling to Rho/Rac GTPase activation, influencing cell differentiation and proliferation. Elevated levels of TEM4, TEM5, TEM6, TEM7, and TEM7R have been observed in breast cancer tissues, suggesting potential therapeutic relevance.
