Target Information

Calcineurin is a Ser/Thr, calcium and calmodulin-dependent protein phosphatase essential in T cell activation. It consists of two subunits:

• Calcineurin A (CnA): 60 kDa catalytic subunit, homologous to protein phosphatases 1 and 2A
• Calcineurin B (CnB): 19 kDa regulatory subunit with four EF-hand calcium binding sites

Calcineurin-mediated dephosphorylation of NF-AT is crucial for its activation and nuclear translocation in T-cells. Immunosuppressive drugs such as cyclosporin A and FK506 inhibit NF-AT activation by binding to immunophilins (cyclophilin and FKBP), forming complexes that block substrate access to CnA’s catalytic domain.
