Description:
The G8.8 monoclonal antibody recognizes the 40 kDa mouse EpCAM (epithelial cellular adhesion molecule), also known as EGP40, 17-1A antigen, TACSTD1, and CD326. The immunogen used was the TE-71 thymic epithelial cell line. CD326 is expressed on most epithelial cells and is considered a pan-carcinoma antigen. It mediates calcium-independent, homophilic cell-cell adhesion and may function as a growth factor receptor. The antigen is being investigated as a target for immunotherapy of human carcinomas. CD326 binds LAIR-1 (CD305) and LAIR-2 (CD306) to inhibit cellular activation and inflammation, playing an important role in tumor biology.

Applications Tested:
Tested by flow cytometric analysis of the TE-71 cell line. Recommended use ≤0.125 µg/test (100 µL final volume). Optimal concentration should be titrated empirically for each assay (10⁵–10⁸ cells/test).
Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.
Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.
Excitation / Emission / Laser:
488–561 nm excitation; 667 nm emission; compatible with Blue, Green, and Yellow-Green lasers.
Target Information
Ep-CAM (epithelial adhesion molecule, ESA) is a 40 kDa transmembrane glycoprotein expressed in epithelial tissues. It functions as a calcium-independent homotypic adhesion molecule and influences cell cycle, proliferation, and metabolism by inducing proto-oncogene c-myc and cyclins A/E. Ep-CAM overexpression correlates with enhanced epithelial proliferation and is highly expressed in human carcinomas, serving as a marker for epithelial-lineage tumors. It localizes to the basolateral surface of most simple epithelia and distinguishes adenocarcinomas from pleural mesotheliomas.
