Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat FOXP3 (Forkhead box P3, also known as SCURFIN or JM2). FOXP3 is a 49–55 kDa transcriptional regulator essential for regulatory T cell (Treg) function. High expression of FOXP3 mRNA is characteristic of CD4⁺CD25⁺ Treg cells, and ectopic expression in CD4⁺CD25⁻ cells induces a Treg phenotype.

Epitope mapping identifies amino acids 75–125 of mouse FOXP3 as the FJK-16s binding region. In humans, this region is alternatively spliced at the mRNA level. Both spliced and non-spliced isoforms are present in CD4⁺CD25⁺ lymphocytes, while mouse splenocytes show only the non-spliced form.

Applications Reported: Intracellular flow cytometric analysis.
Applications Tested: Tested using mouse splenocytes with the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00). Recommended ≤1 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.
Excitation / Emission: 488–561 nm / 667 nm; compatible with Blue, Green, and Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator crucial for immune homeostasis. It is stably expressed in CD25⁺CD4⁺ regulatory T cells and absent in CD8⁺ T cells. FOXP3 mutations cause the scurfy phenotype in mice and IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked) in humans. Transcript variants encoding different isoforms have been identified, and FOXP3 expression in human breast and colon cancer cells is regulated by p53 in response to DNA damage.
