Target Information
CIITA contains an acidic transcriptional activation domain, four LRRs (leucine-rich repeats), and a GTP binding domain. It is located in the nucleus and acts as a positive regulator of class II major histocompatibility complex (MHC II) gene transcription, referred to as the "master control factor" for these genes. CIITA binds GTP to facilitate nuclear transport and functions as a coactivator through intrinsic acetyltransferase activity rather than direct DNA binding.
Mutations in this gene are associated with bare lymphocyte syndrome type II (hereditary MHC class II deficiency), increased susceptibility to rheumatoid arthritis, multiple sclerosis, and possibly myocardial infarction.
