Product Specific Information
The eBioC9B7W monoclonal antibody recognizes mouse CD223 (LAG-3, LAG3) protein expressed by activated α/β-TCR bearing T cells, a subset of γ/δ-TCR bearing T cells, and NK cells.
CD223 is a 70 kDa type I transmembrane protein similar to CD4. In mice, proteolytic cleavage in the D4 domain produces a 54 kDa extracellular fragment and a 16 kDa intracellular fragment. The 54 kDa fragment can remain membrane-associated or be released as soluble CD223.
CD223 binds to MHC class II with higher affinity than CD4, contributing to negative regulation of T-cell activation and homeostatic proliferation. It is also expressed on CD4+CD25+ regulatory T cells and may play a role in their regulatory function.

Applications Tested: Tested on stimulated mouse splenocytes at ≤0.5 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.
Careful titration recommended for optimal performance.

Target Information
LAG-3 is a 70-kDa surface glycoprotein of the Ig superfamily with homology to CD4.
It binds MHC class II with higher affinity than CD4 and negatively regulates T-cell activation and proliferation.
Surface expression is observed on activated T cells (including regulatory T cells) and NK cells.
CD8+ T cells typically express higher LAG-3 levels than CD4+ T cells.
Coexpression of LAG-3 and CD49b identifies Type 1 regulatory T cells (Tr1) in both human and mouse.
