Target Information
Prolyl hydroxylase 4 (P4HTM) is an enzyme that modifies HIF-alpha, playing a critical role in oxygen homeostasis. Unlike classic prolyl hydroxylases that modify collagen in the endoplasmic reticulum, P4HTM acts intracellularly on HIF proteins. Under normoxic conditions, prolyl hydroxylases hydroxylate HIF-alpha, leading to its degradation via the von Hippel-Lindau ubiquitylation complex. Under hypoxic conditions, this hydroxylation is reduced, stabilizing HIF and promoting angiogenesis. P4HTM may therefore function as a cellular oxygen sensor involved in regulating HIF stability.
