Target Information

Peroxiredoxin (Prx) is a growing peroxidase family whose mammalian members are involved in cell proliferation, differentiation, and apoptosis. Many isoforms (about 50 proteins) are grouped according to amino acid sequence homology, active site cysteine residues, and thiol-specific antioxidant activity, and are distributed across all kingdoms.

Mammalian Prx consists of six different members categorized into typical 2-Cys, atypical 2-Cys, and 1-Cys Prx. Except for Prx VI, which belongs to the 1-Cys Prx subgroup, the other five 2-Cys Prx isoforms exhibit thioredoxin-dependent peroxidase (TPx) activity using thioredoxin, thioredoxin reductase, and NADPH as a reducing system.

Mammalian Prxs are 20–30 kDa in molecular size and vary in subcellular localization:

• Prx I, II, and VI: cytosol
• Prx III: mitochondria
• Prx IV: endoplasmic reticulum and secretion
• Prx V: peroxisome, mitochondria, and cytosol
