Store vial at -20°C prior to opening. Aliquot contents and freeze at -20°C or below for extended storage. Avoid repeated freeze-thaw cycles. Centrifuge product if not completely clear after standing at room temperature. The product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

This antiserum is directed against human Thymidylate Synthase and is useful for detecting its presence in various assays. Inhibition of Thymidylate Synthase prevents DNA synthesis and cell proliferation, making it a key target for cancer chemotherapeutic drugs such as fluoropyrimidines used in solid tumor treatment. This antibody detects antigen in human cells and tissues, as well as in bacteria, African green monkey, rat, and mouse. Lower dilutions may be required for some non-human cell lines. Anti-Thymidylate Synthase stains proliferating cells but not nonproliferating cells. Normal colon mucosa shows weak staining, while colorectal cancer specimens often display strong staining.

Target Information

Thymidylate Synthase (TS) catalyzes the conversion of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), essential for DNA synthesis. TS is a major target of fluoropyrimidine-based antineoplastic drugs used in treating solid tumors. In tumor cells, 5-FU and fluorodeoxyuridine are converted to FdUMP, which inactivates TS via formation of a ternary complex with 5,10-methylenetetrahydrofolate. TS expression correlates with response to 5-fluorouracil (5-FU) in colorectal, gastric, head and neck, and breast carcinomas.
