Product Specific Information
- MA1-2022 was used in IHC to detect H2A.X pSer140 in postnatal mouse lung section.
- Detects human and mouse phosphorylated H2AX.
- Successfully used in Western blot, immunofluorescence, and ELISA procedures.
- In Western blot, detects ~17 kDa protein representing phosphorylated H2AX in gamma-irradiated HeLa cell lysate.
- In immunofluorescence, recognizes phosphorylated H2AX in gamma-irradiated HeLa cells.
- Originally validated as part of Thermo Scientific Cellomics High Content Screening Kit; separate antibody may require optimization for best results.
Target Information
Histone H2A.X (H2AX) is a member of the histone H2A family, one of four core histones forming the nucleosome core particle.
DNA double-strand breaks trigger phosphorylation of serine 139 at the C-terminus of H2AX, forming gamma-H2AX, a marker for DNA damage.
Phosphorylation can be detected by Western blot or immunofluorescence to assess DSB frequency.
PI3-kinases, including ATM, DNA-PK, and ATR, are implicated in H2AX phosphorylation.
H2A.X consists of 143 amino acids and is synthesized in G1 and S phases. Its mRNA exists in two forms: one with polyA tail (~1600 bases) and one without (~575 bases).
Histone H2A.X maps to human chromosome 11q23.2–q23.3 and contributes to chromatin structure and DNA repair processes.
