Product Specific Information

MA1-2023 detects human and mouse TIF1 beta.
It has been used successfully in Western blot and immunofluorescence procedures.
By Western blot, this antibody detects a ~100 kDa protein representing TIF1 beta in gamma-irradiated HeLa cell lysate.
In immunofluorescence, MA1-2023 recognizes TIF1 beta in gamma-irradiated HeLa cells.
The immunogen is a recombinant protein corresponding to residues 60–383 of the human TIF1 beta protein.


Target Information

The KRAB (Kruppel-associated box) domain (~45 amino acids) functions as a transcriptional repression domain found in numerous transcription factors.
Over 220 KRAB-zinc finger protein (KRAB-ZFP) genes exist in the human genome. These repress transcription via interaction with KAP-1 (KRAB-associated protein 1), an 835 amino acid polypeptide containing RING finger, B boxes, and a PHD finger.
KAP-1 forms complexes with KRAB-domain transcription factors, enhancing repression efficiency, and interacts directly with HP1 (heterochromatin protein 1) and KRAZ1.
KAP-1 targets KRAZ1 to centromeric heterochromatin foci containing HP1alpha, thereby regulating transcriptional repression.
Mutations that disrupt HP1 binding lead to random KRAZ1 distribution and strong transcriptional activation.
