This antibody reacts with an extracellular epitope of CD11a (alpha-subunit of murine LFA-1), a 180 kDa type I transmembrane glycoprotein expressed on B and T lymphocytes, monocytes, macrophages, neutrophils, basophils, and eosinophils.
Target Information
CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the αLβ2 integrin. CD11a is implicated in activation of the LFA-1 complex and plays a central role in leukocyte intercellular adhesion through interactions with ICAMs 1–3. It also functions in lymphocyte costimulatory signaling and is expressed on the plasma membrane of leukocytes in a low-affinity conformation. Upon cell stimulation, CD11a transitions to a high-affinity state supporting tight ligand binding. CD11a participates in immune cell interaction, tissue-specific settlement, and regulation of T-cell effector functions. Blocking CD11a function is a therapeutic approach in inflammatory diseases; for example, humanized anti-CD11a antibody Efalizumab (Raptiva) interferes with T-cell migration, and simvastatin binding to CD11a modulates immune activity. Two transcript variants encoding different isoforms of CD11a have been identified. Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
